Pilot Investigation on Basal Expression Level of Pro-Inflammatory Mediators in Culture Supernatants or Tissue Lysates of a Reconstructed Human Epidermis Developed in Korea

Abstract
Keraskin¢â-VM is a reconstructed human epidermis developed in Korea. Considering its use for development of skin sensitization or irritation alternative methods, basal expression level of cytokines or chemokines associated with inflammatory signals in skin was investigated. Culture supernatants of Keraskin¢â-VM incubated for 24 h or 48 h were used for determining the level of interleukin (IL)-1¥á, IL-6, IL-18, tumor necrosis factor(TNF)-¥á, monocyte chemotactic protein(MCP)-1, granulocyte macrophage colony-stimulating factor (GM-CSF), macrophage inflammatory protein(MIP)-1¥â, regulated on activation, normal T cell expressed and secreted(RANTES), or vascular endothelial growth factor(VEGF). Tissue lysates were obtained at 48 h. Key mediators for skin inflammation including IL-1¥á, IL-6, IL-18, and TNF¥á were well expressed both in the supernatants and the lysates. Except IL-18, the 24 h supernatants demonstrated higher expression levels than the 48 h supernatants. Major chemokines in the skin such as MCP-1, RANTES, and VEGF were more produced at 48 h than 24 h, wherein GM-CSF and MIP-1¥â were barely detected. Overall, Keraskin¢â-VM could be a valuable reconstructed human epidermis for developing skin sensitization alternative test method.
